VDJ recombination, also known as antigen receptor gene rearrangement or antigen-independent diversification, is a diversity generating assembly process affecting the variable domain of immunoglobulin and TCR genes. : 12/23 rule : B cells : class-switch recombination : CSR : double strand breaks : E2A encoded proteins : hairpin : HMG-1, HMG-2 : lymphoid-specific components : nonlymphoid-restricted components : RAG1 & RAG2 : RSS : SAGA : SHM : somatic hypermutation : synaptic complex

Learn faster with spaced repetition. Three types of CSSR recombination In all the cases red DNA segment is rearranged or moved during recombination ,A,B,X,Y are genes lie in segments of DNA 11. Fig: Mechanism of the site specific recombination by the Cre recombinase 12. VDJ recombination the process by which T cells and B cells randomly assemble different gene segments - known as variable (V), diversity (D) and joining (J) genes - in order to generate unique receptors (known as antigen receptors) that can collectively recognize many different types of molecule The V(D)J recombination is initiated by the dimeric (RAG1-RAG2) 2 complex which specifically recognizes a pair of recombination signal sequences, combinatorially pairs gene segments, and presents hairpin gene segments for opening and ligation by DNA damage repair machineries. Moreover, in contrast to other NHEJxP53 double KO mice, the crossing of TP53 null allele on XLF/Cernunnos KO mice does not result in the onset of pre-B cell malignancies known to emerge from faulty V(D)J recombination.

These segments recombine randomly in a process called VDJ recombination to produce a unique variable domain in the immunoglobulin of each individual B cell. Similar rearrangements occur for the light-chain V region but with only two segments involved: V and J. The theoretical basis of clonal selection is the assumption that lymphocytes bearing an antigen receptor for an antigen exist long before antigen presentation occurs, explained by the idea of random mutations (VDJ recombination) that occur during lymphocyte maturation. During antigen presentation, pre-existing lymphocytes that bear that antigen receptor are merely selected because they can bind with that antigen.

V(D)J Recombination: A Cut-and-Paste Reaction. In the first part of the “cut-and-paste” reaction, breaks within both strands of the DNA helix (double-stranded breaks) are made within the RSS sites; in the second part, the newly created breaks are repaired by the cell's general DNA repair pathway.

• s + v = two mutant alleles and one wildtype allele. • + + + = all wildtype alleles.

In this video lecture we will studyV(D)J RecombinationWe will illustrate V(D)J Recombination in Light chain locus.

in Biochemistry and Molecular Biology from the Institute of Biophysics, Chinese Academy of Sciences in 2012. During his Ph.D. training, he mainly focused on mechanistic understanding of innate immune responses against pathogen infections, especially on the molecular mechanism of DNA recognition by cytosolic double-stranded DNA (dsDNA) sensors as 2003-10-13 2018-11-12 let's now jump into understanding meiosis in some depth so let's start with the germ cell as we mentioned already a germ cell is a cell that it can either go through mitosis to produce other germ cells or it can undergo meiosis in order to produce gametes so this is a germ cell right over here let me draw the nuclear membrane so that's the nuclei let me draw the nucleus a little bit larger Practice: Genetic recombination and conditional knockouts Practice: Inherited disorders: Duchenne muscular dystrophy Practice: Brown adipose tissue and metabolism VDJ recombination would be discussed in details in this class.

VDJ rearrangement on ‘H’ chain occurs in Pro-B cells to produce Heavy chain.

Created by Sal Khan.Watch the next lesson: https://www.khanacademy.org VJ recombination is the mechanism of somatic recombination that occurs only in developing lymphocytes during the early stages of T and B cell maturation. It results in the highly diverse repertoire of antibodies/immunoglobulins and T cell receptors found in B cells and T cells, respectively. The process is a defining feature of the adaptive immune system.

To enhance both the ability of immunoglobulins to recognize and bind to foreign antigens and the effector capacities of the expressed antibodies, naive B cells will undergo class switching recombination (CSR) and somatic hypermutation (SHM). However, the genetics mechanisms of V(D)J recombination, CSR and SHM are not clear.
